The History of Albumin

The word "albumin" traces back to the Latin "albus," meaning white, originally used to describe egg white long before its chemical nature was understood. French chemist Antoine François de Fourcroy formally classified a group of substances he called "albuminous," including egg white and blood serum, in the late 1780s, marking one of the earliest attempts to organize biological materials by shared chemical behavior. Decades later, Dutch chemist Gerardus Johannes Mulder proposed the broader concept of "protein" in 1838, a term Swedish chemist Jöns Jacob Berzelius helped popularize, placing albumin at the very center of the new science of protein chemistry that would eventually explain its role in the human body. By the early 20th century, physicians had already connected low albumin levels to visible swelling and poor nutrition, long before the underlying biochemistry was fully worked out.

What Albumin Actually Is

Albumin is the single most abundant protein circulating in blood plasma, manufactured continuously by the liver and released into the bloodstream. Its primary physical job is maintaining oncotic pressure, the force that keeps fluid inside blood vessels rather than leaking into surrounding tissue — which is why low albumin often causes visible swelling. Beyond that structural role, albumin acts as a transport vehicle, binding to and carrying hormones, vitamins, fatty acids, and many medications through the bloodstream to where they're needed, effectively acting as the blood's general-purpose delivery system. Because it's produced exclusively by the liver and broken down at a fairly predictable rate, albumin levels also serve as a practical window into both liver function and overall nutritional status, with chronically low levels often reflecting long-term rather than sudden changes in health.

How Albumin Is Measured

Most clinical laboratories measure albumin using dye-binding methods, most commonly bromocresol green, which changes color when it binds to albumin in a blood sample, allowing the concentration to be calculated from how much light the resulting solution absorbs. A related dye, bromocresol purple, binds more selectively to albumin and is often preferred for critically ill patients, where bromocresol green can sometimes overestimate results. A separate technique, serum protein electrophoresis, physically separates albumin from the various globulin proteins by passing an electrical current through the sample, giving a visual and quantitative picture of albumin's proportion relative to the rest of the blood's protein content, and revealing patterns useful for diagnosing certain chronic diseases.

Antoine François de Fourcroy's late-18th-century classification work laid the earliest groundwork for recognizing albumin as a distinct biological substance, well before proteins were chemically understood. Gerardus Johannes Mulder's 1838 protein theory, developed with input from Jöns Jacob Berzelius, gave scientists the conceptual framework needed to eventually isolate and study albumin as one specific protein among many. In the modern era, biochemist Theodore Peters Jr. spent much of his career studying serum albumin's structure and function, culminating in his definitive 1996 monograph "All About Albumin," which remains a foundational reference synthesizing more than a century of accumulated research into a single comprehensive account still cited by laboratory scientists today.
